Protein-DNA complexes: Lac Repressor-DNA

David Auble

Images derived from 1lbi (Lac repressor), 1lbh (Lac repressor bound to IPTG), and 1lbg (Lac repressor-DNA complex).

The two windows below allow you to compare the structures of Lac repressor alone and when bound to DNA. The backbone model of the apo form (upper window) is formatted for presentation similarly to the DNA-bound form of Lac repressor shown in the lower window. If you rotate the structures to orient them similarly, you'll notice that the DNA binding helix-turn-helix (HTH) domains of Lac repressor are not visible in the apo structure, presumably because they are disordered. DNA binding by Lac repressor results in the acquisition of HTH domain structure, which is clearly seen as the 'feet' that interact with Lac repressor operator DNA (DNA is shown in orange in the lower window).
