
Purity and quantitation of the DNA template and primer dictate the quality of the sequencing results. All templates intended for automated DNA sequencing should be carefully purified and quantified before being submitted for sequencing. Our current DNA sequencing protocol employs BigDye Terminator labeling chemistry with AmpliTaq polymerase, FS. Since we intend to use a biorobot to handle the reaction set up, we request from our users twice the amounts that actually go into the reaction. Another benefit of keeping a spare aliquot of samples is that, in case we would like to make a rerun under some circumstances, the leftover will be available immediately to avoid any delay.
Our current sample requirements are as follows:
The amount of primer required for the above is exclusively 8-12 picomole. Therefore,
If you use your own primers -- mix the template and primer at the required amount and bring the volume to 14 ul with di. water or dilute buffer; or,
If you like to use the bank primer -- provide the template at the required amount and bring the final volume to 10 UL, with DI water or dilute buffer.
